Field of Research
Biochemistry Department has a long history in the field of molecular biology since 1981, when Prof. Dr. Mohamed Ali Saber started to establish the molecular biology trends and facilities in biochemistry lab.
Different trends of research have been adopted as follows:
1- Schistosomiasis research:
· Biochemical and molecular level: Including host and parasite metabolism, molecular events study, recombinant and DNA vaccine development and genome characterization.
2- Liver research:
· Including molecular studies using in situ hybridization techniques and studies in liver cells culturing and transplantation.
3-Molecular diagnosis and characterization of infectious pathogens:
· Molecular characterization of multi drug resistant bacteria and development of real time PCR assays and kits (Including HCV, HBV, HPV, CMV, H. Pylori etc.)
4- Response to treatment:
· interferon treatment studies for hepatitis C Virus & other pathogens
· Development of lab scale expression and production for selected therapeutic proteins (interferon alpha 2b, interferon gamma, IL12, growth hormone and granulocyte colony stimulating factor).
· Optimization of R& D for pilot scale production of recombinant proteins.
· Designing, bio conjugation and characterization of Nano- and micro-carrier systems (colloidal nanoparticles, polyelectrolyte multilayer capsules, etc) for controlled delivery of therapeutic drugs and genetic materials into targeted cells.
The utilization of nanoparticles to develop novel diagnostic tests for detection of biomarkers of disease and pathogens and other bio-applications.
1. Certain biochemical, pathophysiological and molecular studies in schistosomiasis have been performed on both parasite and host-liver.
2. Development of specific in-situ hybridization techniques, for detection of specific DNA/RNA sequences in liver tissues.
3. Molecular diagnostic and epidemiological studies on hepatitis viruses (B, C and G), Sin virus, papilloma virus, CMV, EBV.
4. Molecular diagnostic and epidemiological studies on pathogens (Salmonella, Helicobacter pylori and Listeria etc.).
5. Molecular Studies on response of HCV infected patients to treatment using recombinant interferon alfa.
6. Biochemical, Epidemiological and Molecular studies of HCV in Egypt.
7. Isolation and molecular identification of four schistosome genes (aldolase, fimbrin, Sm21.7, glucose phosphate isomerase,) (www.ncbi.nlm.nih.gov). These genes have been evaluated as schistosoma vaccine candidates as proteins and as DNA vaccines.
8. Construction of Schistosoma mansoni expression libraries of different life cycle stages which have been used by several countries (distributed internationally through the "Schistosome Geneome Project Network",
9. Isolation and identification of Schistosoma mansoni ESTS through contribution in Schistosome genome project (Saber MA,
www.ncbi.nlm.nih.gov). More than 1650 Scistosoma ESTs are deposited in the GenBank of the NCBI.
10. Hepatocytes isolation and transplantation into experimental animals, either syngenic or xenogenic transplantation. Hepatocytes were immuno-modulated or immuno-isolated and transplanted intrasplenic or intraperitoneal, after induction of acute hepatic failure in transplanted animals.
11. Identification of new mutations in the determinant region of the hepatitis B virus genome isolated from Egyptian patients negative for HBsAb.
12. In collaboration with VACSERA, a construction of recombinant DNA vaccine for the Egyptian strain of foot and mouth disease virus has been developed.
13. Molecular studies on praziquantel resistant Schistosoma mansoni strains.
14. Lab scale production of human interferon alfa 2b, Interferon gamma, growth hormone, IL2 and granulocyte colony stimulating factor.
15. Bacteriological and molecular study of multi drug resistant Mycobacterium tuberculosis in Egypt.

in different E. coli systems, shake flask culture and fermentation, protein folding, one and 2 D protein electrophoresis, protein purification using chromatography and quantification, HPLC analysis, preparative isoelectric focusing and activity assays.
Development of specific polyvalent antibodies, sera treatment, IgG purification, expression libraries immuno-screening, Western blot, dot blot, ELISA, immunohistochemistry and immuno-affinity chromatography.
-In-vitro and In-vivo culturing of hepatocytes:
-Hepatocytes culture, immuno-modulation and transplantation (in spleen and peritoneum).
-MDBK and VERO Cells, and vesicular stomatitis virus (VSV) for antiviral assays.
Retrieve and analysis of DNA and protein sequences, data bases search, primer design, notification of unknown sequences, design of gene models for recombinant protein expression in different host cells.

- In situ hybridization, reverse hybridization, array hybridization.
Synthesis and characterization of nano and micro capsules for controlled delivery of therapeutic drugs and genetic materials into targeted cells (synthesis of capsules, coating, functionalization, cargo loading, encapsulation efficiency, cargo release, cellular uptake, Investigation of the cytotoxic effects the capsules on cells, plasma stability, etc ).
For detection of biomarkers of diseases and pathogens (NPs synthesis, characterization and Functionalization, NPs conjugation with oligonucleotide probes).
Laboratory Structure and Facilities
In addition to the highly experienced staff, Biochemistry and Molecular Biology Department at Theodor Bilharz Research Institute is a well equipped molecular biology and genetic engineering laboratory with well established domains and equipment for different specific fields such as, genetic material isolation, PCR, cloning, cell culturing, protein expression, protein purification, immuno-characterization (ELISA, Western, etc.) and bioinformatics. In 2005 we have established "Biotechnology and Genetic Engineering Unit" for the purpose of R & D in pilot scale production of important proteins especially biopharmaceuticals. This unit has fermentation facilities, protein purification and refolding facilities and tissue culture facilities for biological evaluation of activity of the produced recombinant proteins.NanoBiotechnology unit has been established in 2016 in to promote and support the development of new systems for diagnostics and disease treatment through the research in one of the most promising fields within the new technological advances: the nanoscience.

Biochemistry and Molecular biology Department at TBRI had kept steady record for holding local, national and international training workshops (hands on) in different aspects of molecular biology, biotechnology and Nanobiotecnology as follows:
1. Biochemistry and Molecular Biology Department is considered by WHO as reference training center for African and Arabian countries concerning molecular diagnosis of infectious diseases
2. Training courses in principles of molecular biology.
3. Organizing workshops on molecular forensic diagnosis.
4. Workshops on real time PCR.
5. Workshops on Nanotechniques.
6. Workshops in recombinant protein expression in E. Coli systems.
7. In collaboration with universites, selected graduation research projects have been conducted at Biochemistry and Molecular biology Dept., TBRI.
